ABSTRACT Rheumatoid arthritis, Asthma and acute hypersensitivity are caused by a group of small Ca2+ dependent secretory enzymes called Phospholipase A2 (PLA2's). These enzymes are found in very small amounts or are not present in cells and tissue of normal healthy people, but very large amounts are found in the presence of systemic inflammatory processes. Therefore, a method to inhibit the action or to prevent expression of PLA2 in humans will reduce the incidence of inflammatory processes. A reconstructed cDNA (aplic), 100 % homologous to a domain on the antisense strand of the human APP gene, encodes a 44 amino acid protein (aplip), a potential intracellular activator of PLA2's. Immunotherapeutic substances directed against the protein will block the action of the protein in human cells and tissue. Since `aplip' is not expressed in normal healthy humans the protein is an excellent target for an immunotherapeutic approach to stop rheumatoid arthritis, asthma and other associated diseases in humans, without affecting normal physiological processes in the body.
