The invention provides a method for identifying elements associated with a target molecule comprising the steps of: (a) providing a probe capable of binding by specific molecular interaction to a predetermined specifically defined region of a target molecule, the probe associated with or capable of recruiting an enzyme; (b) adding a tag capable of being activated by the enzyme such that it can attach to elements in the vicinity of the enzyme; and (c) isolating elements having the tag attached thereto, wherein the defined region occurs once, twice, or in a low number of copies in the target molecule. Preferably the tag can attach only to elements in the vicinity of the enzyme.
L'invention se rapporte à un procédé d'identification d'éléments associés à une molécule cible. Ce procédé consiste: à (a) utiliser une sonde pouvant se lier par interaction moléculaire spécifique à une région particulièrement définie et prédéterminée d'une molécule cible, la sonde étant associée à une enzyme ou pouvant en recruter une; à (b) ajouter une étiquette pouvant être activée par ladite enzyme de sorte qu'elle puisse être fixée à des éléments à proximité de l'enzyme; et à (c) isoler les éléments auxquels l'étiquette est fixée. La région définie se reproduit une fois, deux fois ou en nombre réduit de copies dans la molécule cible. De préférence l'étiquette peut se fixer uniquement à des éléments situés à proximité de l'enzyme.
