Derivatives of belactosin and their synthesis are disclosed. In certain embodiments, compounds of the present invention exhibit anti-cancer, antiviral, antibiotic, and/or autoimmune therapeutic abilities. In general, methods of synthesis disclosed herein allow for introduction of a variety of substituents at numerous positions as well as the facile introduction of a beta-lactone ring moiety. The synthetic steps comprise, in preferred embodiments, a tandem Mukaiyama aldol lactonization reaction. Data demonstrating the utility of some of the derivatives as proteasome inhibitors is also disclosed.
La présente invention concerne des dérivés de bélactosine et leur synthèse. Dans certains modes de réalisation, les composés de la présente invention font preuve de propriétés anticancéreuses, antivirales, antibiotiques et/ou de propriétés thérapeutiques autoimmunitaires. En général, les procédés de synthèse présentés dans l'invention permettent l'introduction d'une variété de substituants à de nombreuses positions ainsi que l'introduction aisée d'un fragment d'anneau de bêta-lactone. Les étapes synthétiques comprennent, dans des modes de réalisation préférés, une réaction tandem de lactonisation Mukaiyama aldol.
