A three step method for selection and testing of compounds inhibiting clonal cell growth, consisting of 1) screening for substances that inhibit clonal growth in a culture, 2) in the same culture, testing whether a high local cell concentration (collocation) will decrease the inhibiting effect of such substances on clonal cell growth and 3) testing if export of metastatic cells from a tumour site could be locked by such substances. It should then be possible to decrease or even abolish the development of malignant disease or metastasis from primary tumours and development of benign tumours including atheromas in arteries. The method may also detect compounds that increase clonal growth. These compounds might possess carcinogenic properties or could be used for stimulation of a failing immune system.
La présente invention se rapporte à un procédé en trois étapes de sélection et d'évaluation de composés inhibant la croissance cellulaire clonale. Ledit procédé consiste 1) à cribler des substances qui inhibent la croissance clonale dans une culture, 2) à évaluer, au sein de la même culture, si une concentration cellulaire locale élevée (collocation) peut réduire l'effet inhibiteur de ces substances sur la croissance cellulaire clonale et 3) à évaluer si l'exportation des cellules métastatiques à partir d'un site tumoral peut être bloquée par de telles substances. Il devrait ensuite être possible de réduire ou même de supprimer le développement d'une maladie maligne ou celui de métastases à partir de tumeurs primaires ainsi que le développement de tumeurs bénignes, notamment d'athéromes dans les artères. Le procédé de l'invention peut en outre permettre la détection de composés qui augmentent la croissance clonale. Ces composés peuvent également posséder des propriétés carcinogènes ou être utilisés pour la stimulation d'un système immunitaire déficient.
