An in vivo method for delivering a diagnostic or therapeutic agent to a target site in a mammal, wherein a targeting species including a targeting moiety and a diagnostic or therapeutic agent or a binding site for a subsequently administered diagnostic or therapeutic agent conjugate, the targeting moiety having a primary binding site whereby it specifically binds to the target, is administered and allowed to accrete at the target site, is improved by injecting into the circulatory system of the mammal a clearing agent that binds to the primary binding site of the targeting species, whereby non-localized primary targeting species is cleared from circulation. The method is especially useful in pretargeting methods because the clearing agent does not remove the primary targeting species or block secondary binding sites of the primary targeting species.
L'invention porte sur un procédé in vivo visant à apporter un agent diagnostique ou thérapeutique à un site cible chez un mammifère. Dans le cadre de ce procédé, on apporte à un site cible une espèce ciblante ayant la possibilité de s'accumuler sur celui-ci, l'espèce ciblante comprenant une fraction ciblante et un agent diagnostique ou thérapeutique ou un site de liaison pour un conjugat d'agent diagnostique ou thérapeutique administré ensuite, la fraction ciblante étant pourvue d'un site de liaison primaire au moyen duquel elle se lie de manière spécifique à la cible. On améliore l'espèce ciblante en injectant dans le système circulatoire du mammifère un agent éclaircissant qui se fixe au site primaire de liaison de celle-ci, l'espèce ciblante primaire non localisée étant retirée de la circulation. Ce procédé s'avère particulièrement utile pour des techniques de pré-ciblage puisque l'agent éclaircissant n'élimine pas l'espèce ciblante primaire ni ne bloque des sites secondaires de liaison de celle-ci.
