The invention provides a novel class of compounds (1,2,4-triazolo[4.3-b] pyridazine derivatives) which are ligands for GABAA receptors containing the .alpha.5 subunit, and which satisfy formula (I) wherein R1 and R2 are hydrogen or defined substituents; X is a 5-membered ring containing 1,2,3 or 4 heteroatoms independently chosen from oxygen, nitrogen and sulphur, at most one of the heteroatoms being oxygen or sulphur, or a 6-membered heteroaromatic ring containing 1,2 or 3 nitrogen atoms, the 5- or 6-membered heteroaromatic ring optionally being fused to a benzene ring and optionally being substituted; or X is optionally substituted phenyl; Y is optionally branched C1-4alkylene optionally substituted by an oxo group or Y is a group (CH2)jO wherein the oxygen atom is nearest to group X and j is 2, 3 or 4; and Z is optionally substituted furyl, thienyl or pyridyl; which compounds are useful in the treatment of dementing illness such as Alzheimer's disease.
L'invention concerne une nouvelle classe de composés (dérivés de 1,2,4-triazolo[4,3-b]pyridazine) qui sont de ligands pour les récepteurs GABA¿A? contenant la sous-unité .alpha.5, et qui sont décrits par la formule (I), dans laquelle R?1¿ et R?2¿ sont l'hydrogène ou des substituants définis, X est un cycle formé de 5 éléments contenant 1, 2, 3 ou 4 hétéroatomes sélectionnés indépendamment les uns des autres dans un groupe comprenant l'oxygène, l'azote et le soufre, dans lequel tout au plus un des hétéroatomes est l'oxygène ou le soufre, ou un cycle hétéroaromatique contenant 1, 2 ou 3 atomes d'azote, le cycle hétéroaromatique étant éventuellement condensé avec un cycle benzénique et éventuellement substitué; ou X est le phényle éventuellement substitué; Y est l'alkylène C¿1-4? ramifié éventuellement substitué par un groupe oxo ou Y est un groupe (CH2)¿j?O dans lequel l'atome d'oxygène se trouve à proximité du groupe X et j est 2,3 ou 4; et Z est le furyle, thiényle ou pyridyle substitué. Ces composés sont utiles dans le traitement des démences telles que la maladie d'Alzheimer.
