Peptide inhibitors of serine proteases, especially thrombin, in which the P1- P2 natural amide linkage is replaced by another bond. Exemplary thrombin inhibitors are of the formula: X-(aa3)-(aa2)--(aa1)-Z wherein X is H or a substituent on the N-terminal amino group, aa3 is a hydrophobic amino acid such as Phe, aa2 is Pro, aa1 is Arg or an Arg analogue such as methoxypropylglycyl, Z is -COOH or a heteroatom acid group, such as boronate, or a derivative of either, and is a non-amide linkage, typically containing up to 5 in-chain atoms, such as -CO2-, -CH2O-, NHCO- or -CH2-CH2-.

On décrit des inhibiteurs peptidiques de sérines protéases, notamment la thrombine, dans lesquels la liaison amide naturelle P1-P2 est remplacée par une autre liaison. A titre d'exemple, on décrit des inhibiteurs de thrombine possédant la formule X-(aa?3¿)-(aa?2¿)-?-(aa?1¿)-Z dans laquelle X représente H ou un substituant sur le groupe amino N-terminal, aa?3¿ représente un acide aminé hydrophobe tel que Phe, aa?2¿ représente Pro, aa?1¿ représente Arg ou un analogue de Arg tel que méthoxypropylglycyle, Z représente -COOH ou un groupe acide d'hétéroatomes, tel qu'un boronate, ou un dérivé de l'un ou de l'autre, et ? représente une liaison non amide, contenant typiquement jusqu'à 5 atomes en chaîne, tels que -CO¿2?-, -CH¿2?O-, NHCO- ou -CH¿2?-CH¿2?-.
