The present invention relates to a recombinant herpes simplex virus (HSV) viral vector genome which has substantially lost its transducing properties as a result of a DNA sequence change in the gene coding for Vmw65 protein and also comprises an expressable heterologous gene inserted into a region of the HSV genome which is non-essential for the culture of the virus, the gene being under the control of the immediate early (IE1) gene enhancer of cytomegalovirus (CMV) and to the use of the recombinant HSV genome in therapy and vaccination.

La présente invention se rapporte à un génome vectoriel viral de recombinaison de l'herpèsvirus (HSV) qui a pratiquement perdu ses propriétés de transduction suite à une modification de séquence d'ADN dans le gène codant pour la protéine Vmw65 et qui comporte également un gène hétérologue exprimable inséré dans une région du génome de l'herpèsvirus qui n'est pas essentielle pour la culture du virus, le gène étant sous le contrôle de l'activateur immédiat du gène précoce (IE1) du cytomégalovirus (CMV). L'invention se rapporte également à l'utilisation du génome de recombinaison du HSV en thérapie et dans les vaccinations.
