A method and device for treating a mammalian organism to obtain a desired local or systemic physiological or pharmacological effect is provided. The method includes administering a sustained release drug delivery system to a mammalian organism in need of such treatment at an area wherein release of en effective agent is desired and allowing the effective agent to pass through the device in a controlled manner. The device includes an inner core or reservoir comprising the effective agent; a first coating layer, which is permeable to the passage of the effective agent; a second coating layer, which is essentially impermeable to the passage of the effective agent; and a third coating layer, which is permeable to the passage of the effective agent. The first coating layer covers at least a portion of the inner core. The second coating layer covers at least a portion of the first coating layer and inner core; however, at least a small portion of the first coating layer or inner core is not coated with the second coating layer. The second coating layer includes an impermeable film and at least one impermeable disc. The third coating layer essentially completely covers the second coating layer and the uncoated portion of the first coating layer or inner core.

L'invention a trait à une technique ainsi qu'au dispositif correspondant permettant de traiter un organisme de mammifère et ce, afin d'obtenir un effet pharmacologique ou physiologique souhaité, général ou local. La technique consiste à administrer un système d'administration de médicament à dégagement prolongé à un mammifère nécessitant un tel traitement dans une région où le dégagement d'un agent efficace est souhaité et à faire en sorte que cet agent puisse traverser le dispositif de manière régulée. Le dispositif comporte un noyau interne ou réservoir renfermant l'agent efficace, une première couche d'enrobage, laissant passer cet agent, une deuxième couche d'enrobage interdisant quasiment le passage dudit agent et une troisième couche d'enrobage le laissant passer. La première couche d'enrobage recouvre au moins une partie du noyau interne, la deuxième au moins une partie de la première couche et du noyau interne, une petite partie au moins de la première couche d'enrobage ou du noyau interne n'étant, toutefois, pas recouverte par la deuxième couche d'enrobage. La deuxième couche d'enrobage comprend un film imperméable et au moins un disque imperméable. La troisième couche d'enrobage recouvre quasiment la totalité de la deuxième couche d'enrobage et la partie non revêtue de la première couche d'enrobage ou du noyau interne.
