The invention concerns the use of duplex oligonucleotides about 25 to 30 base pairs to introduce site specific genetic alterations in plant cells. The oligonucleotides can be delivered by mechanical (biolistic) systems or by electroporation of plant protoplasts. Thereafter plants having the genetic alteration can be generated from the altered cells. In specific embodiments the invention concerns alteration in the gene that encodes acid invertase, UDP- glucose pyrophosphorylase, polyphenol oxidase, O-methyl transferase, cinnamyl alcohol dehydrogenase, ACC synthase and ACC oxidase or etr-1 or a homolog of etr-1, and plants having isolated point mutations in such genes.
L'invention concerne l'utilisation d'oligonucléotides à double hélice, environ 25 à 30 paires de bases, pour introduire des modifications génétiques spécifiques à un site dans des cellules végétales. Les oligonucléotides peuvent être introduits par des systèmes mécaniques (biolistiques) ou par électroporation de protoplastes végétaux. Par la suite, des plantes présentant la modification génétique peuvent être générées à partir des cellules modifiées. Dans des modes de réalisation spécifiques, l'invention concerne une modification du gène codant une acide invertase, une UDP-glucose pyrophosphorylase, une polyphénol oxydase, une O-méthyle transférase, une cinnamyl alcool déshydrogénase, une ACC synthase et une ACC oxydase ou etr-1 ou un homologue d'etr-1, et des plantes présentant des mutations ponctuelles isolées dans ces gènes.
