The present invention relates to an isolated promoter region of the mammalian transcription factor FOXC2. The invention also relates to screening methods for agents modulating the expression of FOXC2 and thereby being potentially useful for the treatment of medical conditions related to obesity. The invention further relates to a previously unknown variant of the human FOXC2 gene, derived via the use of an alternative promoter, which produces an additional exon that generates a distinct open reading frame via splicing. The alternative gene encodes a variant of the FOXC2 transcription factor, which is lacking a part of the DNA-binding domain and consequently has a potential regulatory function.
La présente invention concerne une zone de promoteur isolée du facteur de transcription FOXC2 d'un mammifère. L'invention concerne des procédés de sélection permettant à des agents de moduler l'expression de FOXC2 et d'être ainsi potentiellement utiles pour traiter des états médicaux liés à l'obésité. L'invention concerne également une variante précédemment inconnue du gène humain FOXC2, dérivé par l'utilisation d'un promoteur alternatif, qui produit un exon additionnel qui génère un cadre de lecture ouvert distinct par épissage. Le gène alternatif code une variante du facteur de transcription FOXC2 auquel manque une partie du domaine de liaison à l'ADN et, par conséquent, a une fonction de régulation de potentiel.
