There is disclosed an isolated cDNA sequence (SEQ ID NO:1) encoding a P4.33 polypeptide and comprising a coding region (SEQ ID NO:2) of the sequence described in SEQ ID NO:1, or a sequence having at least 90 % homology with the coding region of SEQ ID NO:1. The P4.33 polypeptide functions as a specific cell-surface receptor for IGFBP-3, and undergoes nuclear translocation in combination with IGFBP-3. IGFBP-3 and P4.33 (IGFBP-3R) cooperatively suppress DNA synthesis and cell growth, and induce caspase activation and apoptosis in cancer cells, indicating that P4.33 is an important mediator of IGF- independent growth inhibitory actions of IGFBP-3. The P4.33:IGFBP-3 system of the present invention can be used, inter alia, in screening and diagnostic assays, and for therapeutic methods for cancer treatment and tumor suppression.
L'invention concerne une séquence d'ADNc isolée (SEQ ID NO:1) codant pour un polypeptide de P4.33 et comprenant une région codante (SEQ ID NO:2) de la séquence décrite dans SEQ ID NO:1, ou une séquence présentant une homologie d'au moins 90 % avec la région codante de la séquence (SEQ ID NO:1). Le polypeptide de P4.33 fonctionne comme un récepteur antigénique spécifique pour l'IGFBP-3, et il est soumis à une translocation nucléaire en combinaison avec l'IGFBP-3. L'IGFBP-3 et la P4.33 (IGFBP-3R) ensemble permettent de supprimer la réplication de l'ADN et la croissance cellulaire, et elles déclenchent l'activation de la caspase et l'apoptose dans les cellules cancéreuses; ceci indique que la P4.33 est un médiateur important des actions inhibitrices de la croissance ne dépendant pas de l'IGF du IGFBP-3. L'ensemble P4.33:IGFBP-3 décrit dans cette invention peut être utilisé entre autres pour des méthodes de criblage et diagnostiques, et pour des méthodes thérapeutiques permettant le traitement du cancer et la suppression d'une tumeur.
