Improvements to the polymerase chain reaction (PCR), a process for in vitro enzymatic amplification of specific nucleic acid sequences, can be achieved by changing the way that PCR reagents are mixed and the enzymatic reaction is started and by the replacement of mineral oil, commonly used as a vapor barrier to minimize solvent evaporation, by a grease or wax. The use of such mixtures allows for the delay of reagent mixing until the first heating step of a PCR amplification, thereby reducing the enzymatic generation of nonspecific products which occurs when a complete mixture of PCR reagents, with or without test sample, stands at room temperature or below. These mixtures increase the shelf-life of PCR reagents and increase protection of the laboratory environment against contamination by PCR product.

Des améliorations de l'amplification en chaîne par la polymérase (ACP) - méthode d'amplification enzymatique in vitro de séquences d'acides nucléiques spécifiques - peuvent être obtenues en modifiant la technique de mélange des réactifs ACP ainsi que le démarrage de la réaction enzymatique, et en remplaçant l'huile minérale, généralement employée comme barrière pour la vapeur - réduisant ainsi l'évaporation de solvant - par une graisse ou une cire. L'utilisation de ce type de mélange permet de retarder le mélange des réactifs jusqu'à la première étape de chauffage de l'ACP, réduisant ainsi la production enzymatique de produits non spécifiques, qui a lieu lorsqu'un mélange complet de réactifs ACP, avec ou sans échantillon à analyser, repose à la température de la pièce ou en-dessous. Enfin, ce type de mélange augmente la durée de conservation des réactifs ACP et améliore la protection contre la contamination par les produits ACP en laboratoire.
