A low dose antidiabetic pharmaceutical formulation is provided, especially adapted for treating Type II diabetes in drug naive patients, which includes a combination of metformin (employed in a reduced amount (less than 800 mg metformin per day) compared to that employed in generally accepted medical practice) and at least one other antidiabetic agent such as a sulfonyl urea, for example, glyburide, which combination provides at least about substantially equivalent efficacy in treating diabetes in drug naive patients, as do antidiabetic formulations containing metformin employed in dosages prescribed in generally accepted medical practice for first line therapy in treating diabetes, but with substantially reduced side effects, such as hypoglycemia and/or gastrointestinal distress. A method for treating diabetes in drug naive human patients is also provided employing the above formulation to reduce insulin resistance and/or post-prandial glucose excursion and/or hemoglobin 1Ac, and/or increase post-prandial insulin, thereby treating the diabetes.
L'invention concerne une préparation pharmaceutique antidiabétique à faible dosage, spécialement conçue pour traiter le diabète de type II chez des novices aux essais cliniques. Cette préparation comprend une combinaison de metformine (utilisée en quantité réduite (moins de 800 mg de metformine par jour) par rapport à la quantité utilisée dans la pratique médicale courante) et au moins un autre agent antidiabétique tel que qu'un sulfonylurée, par exemple du glibenclamide. Cette combinaison présente une efficacité au moins sensiblement équivalente pour le traitement du diabète chez des novices aux essais cliniques, aux préparations antidiabétiques contenant de la metformine à des dosages prescrits dans la pratique médicale courante pour une thérapie immédiate destinée au traitement du diabète, mais avec des effets secondaires sensiblement réduits, tels que l'hypoglycémie et/ou les troubles gastro-intestinaux. L'invention concerne également une méthode permettant de traiter le diabète chez des novices aux essais cliniques à l'aide de la préparation susmentionnée afin de réduire l'insulinoresistance et/ou le déplacement postprandial du glucose et/ou l'hémoglobine 1Ac, et/ou d'augmenter l'insuline postprandiale, ce qui permet ainsi de traiter le diabète.
