Plant viral vectors have great potential in rapid production of proteins, but no simple Here a gemillivirus-based system for high-yield and rapid production of oligomeric protein complexes, including virus-like particle (VLP) vaccines and monoclonal antibodies (mAbs) is described. In particular, a single vector that contains two non-competing replicons for transient expression in Nicotiana benthamiana leaves is described. The correct assembly of these subunit proteins into functional oligomer-ic structures (VLPs or full-size mAb) is also described. This system advances plant transient expression technology by eliminating the need for non-competing viruses, and thus, enhances the realistic commercial application of this technology for producing mul-tiple-subunit protein complexes.

Les vecteurs végétaux viraux présentent un gros potentiel en matière de production rapide de protéines, mais rien n'est simple en ce domaine. La présente invention concerne, donc, un système fondé sur un géminivirus permettant une production rapide et à haut rendement de complexes protéiques oligomères, dont des vaccins composés de particules pseudo-virales (VLP) et des anticorps monoclonaux (mAb). La présente invention concerne, en particulier, un vecteur unique comportant deux réplicons non-concurrents en vue d'une expression transitoire dans des feuilles de Nicotiana benthamiana. L'invention concerne également le bon assemblage de ces sous-unités protéiques en structures oligomères fonctionnelles (VLP ou mAb pleine longueur). Ce système constitue une avancée dans la technologie de l'expression transitoire chez les végétaux en évitant le recours à des virus non-concurrents et cela rend donc plus réaliste une application commerciale de cette technologie à des fins de production de complexes protéiques à sous-unités multiples.
