The present invention relates to novel .alpha.-keto carbonyl calpain inhibitors for the treatment of neurodegenerative diseases and neuromuscular diseases including Duchenne Muscular Dystrophy, Becker Muscular Dystrophy and other muscular dystrophies. Disuse atrophy and general muscle wasting can also be treated. Diseases of the eye, in particular cataract, can be treated as well. Generally all conditions where elevated levels of calpains are involved can be treated. The compounds of the invention may also inhibit other thiol proteases such as cathepsin B, cathepsin H, cathepsin L, papain or the like. Multicatalytic Protease (MCP) also known as proteasome may also be inhibited and the compounds can therefore be . used to treat cell proliferative diseases such as cancer, psoriasis, and restenosis. The compounds of the present invention are also inhibitors of cell damage by oxidative stress through free radicals and can be used to treat mitochondrial disorders and neurodegenerative diseases, where elevated levels of oxidative stress are involved.

L'invention concerne de nouveaux inhibiteurs de calpaine de type .alpha.-céto carbonyle, servant à traiter des maladies neurodégénératives et des maladies neuromusculaires parmi lesquelles figurent la dystrophie musculaire de Duchenne, la dystrophie musculaire de Becker, ainsi que d'autres dystrophies musculaires. Ces inhibiteurs permettent également de traiter une atrophie due à l'inactivité, une fonte musculaire générale, des maladies oculaires, notamment une cataracte, et de manière générale, tout état pathologique impliquant des niveaux élevés de calpaine. Les composés selon l'invention peuvent aussi inhiber d'autres thiol-protéases telles que la cathepsine B, la cathepsine H, la cathepsine L, la papaine ou analogue. Par ailleurs, lesdits composés peuvent inhiber la protéase multicatalytique (MCP) encore appelée protéasome et peuvent par conséquent être utilisés pour traiter des maladies liées à une prolifération cellulaire telles que le cancer, le psoriasis et la resténose. Ces composés inhibent en outre les altérations cellulaires induites par le stress oxydatif par l'intermédiaire de radicaux libres, et peuvent être employés pour traiter des troubles mitochondriaux ainsi que des maladies neurogénénératives impliquant des niveaux élevés de stress oxydatif.
