It is intended to provide a novel meiosis-specific centromeric protein Sgo1 (shugoshin) originating in a fission yeast Schizosaccharomyces pombe as a factor ensuring the retention of unidirection and adhesion in sister centromeres in the first meiotic division synergistically with cohesin, its homolog or paralog having an activity of regulating chromosomal distribution, and DNA encoding the same. To clarify a protein protecting Rec8 in the latter stage of division, screening is made of a gene which regulates mitotic division development and inhibits the separation of sister chromatids in the latter stage of division, when expressed together with Rec8, from among fission yeast genes. As the result of this approach, a meiosis-specific protein Sgo1 which protects (shugo) centrometic Rec8 from degradation in the latter stage of first division is found out. Moreover, an Sgo1 homolog of a budding yeast and a mitotic division paralog Sgo2 of a fission yeast are found out.

L'invention concerne une nouvelle protéine centromérique spécifique à la méiose Sgo1 (shugoshine) obtenue de la levure de fission Schizosaccharomyces pombe comme facteur garantissant l'unidirection et l'adhésion des centromères soeurs au cours de la première division méiotique de manière synergétique avec la cohésine, son homologue ou paralogue possédant une activité de régulation de la distribution chromosomique, et un ADN codant pour celle-ci. Pour mettre en évidence une protéine protégeant Rec8 au cours de la dernière phase de division, le criblage d'un gène régulant le développement de la divison mitotique et inhibant la séparation des chromatides soeurs est effectué dans la dernière phase de division, lorsqu'il est exprimé avec Rec8, parmi des gènes de levures de fission. Cette approche permet de définir une protéine Sgo1 spécifique à la méiose protégeant (shugo) Rec8 centromérique contre la dégradation dans la dernière phase de la première division. L'invention concerne, de plus, la découverte d'un homologue Sgo1 d'une levure de bourgeonnement et d'un paralogue de division mitotique Sgo2 d'une levure de fission.
