This invention relates to novel lactams having formula (I) to their pharmaceutical compositions and to their methods of use. These novel compounds inhibit the processing of amyloid precursor protein and, more specifically, inhibit the production of A.beta.-peptide, thereby acting to prevent the formation of neurological deposits of amyloid protein. More particularly, the present invention relates to the treatment of neurological disorders related to .beta.-amyloid production such as Alzheimer's disease and Down's Syndrome.
Cette invention se rapporte à de nouveaux lactames représentés par la formule (I) et à leurs compositions pharmaceutiques et leurs procédés d'utilisation. Ces nouveaux composés empêchent la transformation de la protéine précurseur amyloïde et, plus spécifiquement empêchent la production de peptide A.beta., ayant ainsi une action préventive sur la formation des dépôts neurologiques de protéine amyloïde. Cette invention se rapporte plus particulièrement au traitement des troubles neurologiques associés à la production d'amyloïde .beta., tels que la maladie d'Alzheimer et le syndrome de Down.
