The present inventors successfully constructed expression vectors that enable high-level production of foreign gene-derived proteins in mammalian host cells, which comprise a translation-impaired drug resistance gene cistron whose expression has been attenuated by altering the codons to the least frequently used codons in mammals; and a gene cassette which has a cloning site for incorporation of a foreign gene between a highly transcriptionally active promoter and a highly stable polyadenylation signal.

La présente invention concerne un vecteur d'expression qui comprend une cassette de gènes contenant un gène cistron d'expression réduite résistant à un médicament et qui possède une propriété de détérioration de la traduction et un site de clonage destiné à l'insertion d'un gène étranger entre un promoteur hautement actif transcriptionnellement et un signal de polyadénylation hautement stable. Le vecteur selon l'invention est capable de produire une protéine dérivée d'un gène étranger à un niveau élevé dans une cellule hôte mammalienne.
