This invention relates to methods and compositions useful for delivering antigens to dendritic cells which are then useful for inducing antigen- specific cytotoxic T lymphocytes and T helper cells. This invention also provides assays for evaluating the activity of cytotoxic T lymphocytes. According to the invention, antigens are targeted to dendritic cells by apoptotic cells which may also be modified to express non-native antigens for presentation to the dendritic cells. The dendritic cells which are primed by the apoptotic cells are capable of processing and presenting the processed antigen and inducing cytotoxic T lymphocyte activity or may also be used in vaccine therapies.
L'invention se rapporte à des procédés et à des compositions visant à apporter des antigènes aux cellules dendritiques qui, à leur tour, servent à induire des lymphocytes T cytotoxiques spécifiques à l'antigène et des lymphocytes T auxiliaires. L'invention concerne également des dosages pour évaluer l'activité des lymphocytes T cytotoxiques. Selon cette invention, les antigènes sont dirigés sur les cellules dendritiques par les cellules d'apoptose, qui peuvent être également modifiées pour exprimer les antigènes non natifs, pour présentation aux cellules dendritiques. Les cellules dendritiques, qui utilisent comme amorce les cellules d'apoptose, peuvent effectuer un traitement et présenter l'oxygène traité ainsi qu'induire une activité de lymphocytes T cytotoxiques. Elles peuvent aussi être utilisées dans des thérapies à base de vaccins.
