The invention provides methods and compositions for treating a cancerous condition, using one or more inhibitory agents that target the transcription factors Sp1 or Sp3. Compositions provided include Sp1 and Sp3 - targeted antisense oligonucleotides and ribozymes and recombinant viral vectors for delivery of these ribozymes to a tumor, as well as Sp1/Sp3 specific promoters for optimized negative-feedback expression of the Sp1/Sp3 ribozymes. The invention further provides a system for analyzing the effectiveness of a cancer therapy treatment on a cancer by using an animal model in which the cancerous cells and recombinant cancer therapeutics are separately marked with different fluorescent markers that allow for the detection of effectively treated cancer cells and tissue.

Cette invention se rapporte à des procédés et à des compositions pour traiter un état cancéreux, en utilisant un ou plusieurs agents inhibiteurs qui ciblent les facteurs de transcription Sp1 ou Sp3. Les compositions proposées contiennent des oligonucléotides et des ribozymes anti-sens ciblés sur les facteurs Sp1 et Sp3 et des vecteurs viraux recombinés servant à l'apport de ces ribozymes à la tumeur, ainsi que des promoteurs spécifiques des facteurs Sp1/Sp3 pour l'expression à rétroaction négative optimisée des ribozymes Sp1/Sp3. Cette invention concerne également un système servant à analyser l'efficacité d'un traitement anticancéreux sur un cancer en utilisant un modèle animal dans lequel les cellules cancéreuses et les agents thérapeutiques anticancéreux recombinés sont marqués séparément avec différents marqueurs fluorescents qui permettent la détection des cellules et des tissus cancéreux traités efficacement.
