The invention provides methods, compositions, and kits for fragmentation and labeling of nucleic acids. More particularly, the invention relates to methods for fragmentation of nucleic acids to produce fragments with 3' end hydroxyl groups within a desired size range. In methods of the invention, nucleic acids are fragmented at abasic sites to produce fragments with blocked 3' ends. The 3' ends are unblocked to produce polynucleotide fragments with hydroxyl groups at their 3' ends. Methods, kits, and compositions for carrying out fragmentation of a polynucleotide template in a single reaction mixture to yield fragments with 3'-hydroxl ends within the desired size range are disclosed.

La présente invention concerne des procédés, des compositions et des trousses permettant de fragmenter et de marquer des acides nucléiques. Cette invention porte plus particulièrement sur des procédés de fragmentation d'acides nucléiques servant à produire des fragments ayant des groupes hydroxyle terminaux 3' qui se situent dans une plage de dimensions désirées. Dans les procédés selon l'invention, les acides nucléiques sont fragmentés au niveau de sites abasiques pour produire des fragments ayant les extrémités 3' bloquées. Les extrémités 3' sont débloquées pour produire des fragments polynucléotidiques ayant des groupes hydroxyle au niveau de leurs extrémités 3'. Cette invention concerne également des procédés, des trousses et des compositions permettant d'effectuer la fragmentation d'une matrice polynucléotidique dans un mélange de réaction unique pour produire des fragments à extrémités hydroxyle 3' dont les dimensions se situent dans la plage de dimensions désirées.
