The present invention provides slow release estradiol-progesterone formulations that can be used in either contraception or hormone replacement therapies. The formulations comprise shaped particles of estradiol that is in a hemicrystalline form that exhibits especially low dissolution rates. The shaped particles comprise estradiol compounded in a 1:1 molar ratio with cholesterol, and are administered in combination with progesterone. The slow release formulations of the present invention afford the dual advantages of a low dose estradiol formulation with a low frequency administration regimen. The formulations can be parenterally administered once a month or less often.
La présente invention concerne des formulations d'estradiol-progestérone à libération lente pouvant être utilisées soit en contraception soit pour les traitements hormonaux substitutifs. Ces formulations comprennent des particules d'une forme particulière d'estradiol semi-cristallin qui présente des vitesses de dissolution particulièrement faibles. Lesdites particules comprennent un mélange constitué d'estradiol/cholestérol en un rapport molaire de 1:1, et sont administrées en association avec la progestérone. Les formulations à libération lente selon la présente invention sont doublement avantageuses en ce sens qu'elles présentent une formulation à faible dose d'estradiol et nécessitent une fréquence d'administration peu élevée. Ces formulations peuvent être administrées par voie parentérale une fois par mois ou à intervalles plus longs.
