A method for the extraction of microbial DNA which renders such DNA amenable for further enzymatic modification in situ, including digestion of the DNA with restriction endonuclease. The method comprises: a) heating the microbial cells in the presence of a hypotonic EDTA solution, b) lysing the cells by addition of achromopeptidase and c) heating said lysed cells to inactivate remaining protease and nuclease.

L'invention concerne un procédé d'extraction de l'ADN microbien qui rend cet ADN apte à une modification enzymatique ultérieure in situ, et qui consiste à digérer l'ADN par l'endonucléase de restriction. Le procédé consiste à: a) chauffer les cellules microbiennes en présence d'une solution hypotonique d'EDTA, b) lyser les cellules par addition d'une achromopeptidase et, c) chauffer ces cellules lysées afin d'inactiver la protéase et la nucléase restantes.
