Active recombinant human IL-4 is purified from a crude fermentation broth of a secreted E. coli mutant by subjecting said solution to selective chromatography on a zinc-chelating agarose column at pH 7.2 in a buffer containing a high concentration of sodium chloride, i.e., about 1 M sodium chloride, washing first with a phosphate buffer at pH 7.2 containing 1.0M sodium chloride then with a buffer containing 10% glycerol and eluting at pH 5Ø The resulting purified active recombinant human IL-4 solution is further treated by ration exchange chromatography, concentration by diafiltation up to 20 mg/mL and size exclusion gel chromatography using a citrate buffer at pH 4.5.
On purifie de l'IL-4 humaine recombinante active à partir d'un bouillon de fermentation brute d'un mutant E. coli sécrété en soumettant ladite solution à la chromatographie sélective sur une colonne d'agarose chélateur du zinc ayant un pH de 7,2 dans un tampon contenant une forte concentration de chlorure de sodium, c'est-à-dire environ 1M de chlorure de sodium, en procédant à un premier lavage avec un tampon de phosphate ayant un pH de 7,2 et contenant 1,0M de chlorure de sodium, puis à un lavage avec un tampon contenant 10 % de glycérine et en éluant à un pH de 5,0. La solution d'IL-4 humaine recombinante active purifiée ainsi obtenue est en outre traitée par chromatographie avec échange cationique, par concentration par diafiltration jusqu'à 20mg/ml et par chromatographie sur gel à exclusion granulométrique utilisant un tampon de citrate ayant un pH de 4,5.
