Novel recombinant anticoagulation proteins, methods of their use and methods of their production are described. In particular, recombinant fusions of annexin V (ANV) and Kunitz protease inhibitors (KPI) that possess potent anticoagulant activity are provided. The fusions, abbreviated ANV:KPI, utilize ANV having high affinity for phosphatidyl-L-serine with various KPI's to target serine proteases in membrane-associated coagulation complexes in the blood coagulation cascade. ANV:KPIs are potentially useful antithrombotic drugs permitting localized passivation of thrombogenic vessel walls and associated thrombi.
La présente invention concerne de nouvelles protéines de recombinaison anticoagulantes, des techniques d'utilisation et des techniques de production de ces protéines. Cette invention concerne, en particulier, des fusions recombinantes d'annexine V (ANV) et d'inhibiteurs de protéase de type Kunitz qui possèdent une activité anticoagulante puissante. Ces fusions, abrégées en ANV: KPI, utilise une ANV possédant une grande affinité pour la phosphatidyl-Lsérine avec divers KPI de façon à cibler des sérine protéases dans des complexes de coagulation associés à la membrane dans les réactions en cascade de la coagulation du sang. Ces fusions ANV: KPI sont potentiellement utiles comme médicaments antithrombotiques permettant une <=passivation>= localisée des parois des vaisseaux thrombotiques et de thrombi associés.
