The present invention describes an improved method of generation of Brassica transformants based on a method of selecting genetically transformed Brassica juncea explants based on their ability to utilize Xylose as a sole carbohydrate source. The said invention encompasses the process of Agrobacterium Mediated Transformation of the target host plant with the constructed vector for the expression of the enzyme Xylose Isomerase. Also disclosed is the method of selecting the putative transformants subsequent to transformation with the said vector to obtain a metabolic advantage of utilizing Xylose as a carbon source. The subject invention alleviates the disadvantage of the negative selection methods. Reported is a stable transformation efficiency of 34-40% using Xylose Isomerase for selection.
La présente invention concerne un procédé amélioré pour produire des transformants de Brassica sur la base d'une méthode de sélection d'explants de Brassica juncea génétiquement transformés en fonction de leur capacité à utiliser le xylose comme source unique d'hydrates de carbone. L'invention concerne un procédé de transformation, médié par une agrobactérie, de la plante hôte cible avec le vecteur assemblé pour l'expression de l'enzyme xylose isomérase. Elle concerne également un procédé de sélection des transformants putatifs après transformation avec ledit vecteur afin d'obtenir l'avantage métabolique résultant de l'utilisation de xylose comme source de carbone. La présente invention atténue les inconvénients des méthodes de sélection négative. Elle permet d'atteindre un rendement de transformation stable de 34 à 40 % en utilisant la xylose isomérase pour la sélection.
