The present invention relates to novel colchicine derivatives of formula (1), where Y is a -CH2-CH-NH-R3 group or a -CH-CH2OR3 group and the other variables are as defined in claim 1, having antiproliferative, antitumoral and anti- inflammatory activities. The novel compounds have a cytotoxicity on human tumoral cell lines comparable with colchicine, but, in comparison with the latter, they are less toxic and more selective, particularly on cells resistant to the usual medicaments. Some compounds have a marked activity on TNF and interleukine 2, and therefore are very potent anti-inflammatory agents. They can be included in pharmaceutical formulations useful for the parenteral, oral and topical administrations.

On décrit de nouveaux dérivés de la colchicine de la formule (1) dans laquelle Y représente un groupe -CH¿2?-CH-NH-R¿3? ou un groupe -CH-CH¿2?OR¿3? et les autres variables ont les notations données dans la revendication 1, ces dérivés possédant des activités antiprolifératives, antitumorales et anti-inflammatoires. Les nouveaux composés présentent une cytotoxicité sur les lignées cellulaires tumorales humaines qui est comparable à celle de la colchicine, mais ils sont, par rapport à cette substance, moins toxiques et plus sélectifs, notamment à l'égard des cellules résistant aux médicaments usuels. Certains composés possèdent une activité marquée sur le facteur de nécrose tumorale et sur l'interleukine 2 et constituent, par conséquent, des agents anti-inflammatoires très puissants. On peut inclure ces composés dans des formulations pharmaceutiques utiles à une administration parentérale, orale et topique.
