Disclosed are a variety of peptide conjugates represented by the following general formula (I): R1-Z-X-Z'-R2, wherein X represents a hexapeptide of the formula (II): A1-A2-A3-A4-A5-A6 wherein A1 represents Arg, Lys, or His, A2 represents Tyr, Trp, or Phe, A3 represents Tyr, Asn, Trp or Phe, A4 represents Lys, Arg or His, A5 represents Phe, Tyr, Trp, Leu, Val or Ile, and A6 represents Arg, Lys, or His and wherein each amino acid residue in said hexapeptide may be in the L or D form; Z represents a charged peptide chain of from 4 to 20 amino acid residues having the D or L configuration or is missing; and Z' represents a charged peptide chain of from 4 to 20 amino acid residues having the D or L configuration or is missing, providing that not both of Z and Z' are missing; R1 represents H or an acyl group; R2 represents NR3R4 where each of R3 and R4 independently represents hydrogen, C(1-6)alkoxy, aryloxy, or a lower alkyl as defined herein; or R2 represents OH; the peptide conjugates of formula (I) being optionally further linked to a transport moiety; and salts, hydrates and solvates thereof, and C-terminally amidated or esterified derivatives thereof with suitable organic or inorganic acids, including methods or making and using such conjugates. Also provided are antibodies that specifically bind the peptide conjugates. The present invention has a wide spectrum of important applications including use in the treatment of disorders impacted by nociceptin and related opioid-like peptides.
Cette invention a trait à divers conjugats peptidiques correspondant à la formule I, R¿1?-Z-X-Z'-R¿2?, formule dans laquelle X représente un hexapeptide correspondant à la formule II, A?1¿-A?2¿-A?3¿-A?4¿-A?5¿-A?6¿. Dans cette formule, A?1¿ représente Arg, Lys ou His, A?2¿ représente Tyr, Trp Phe, A?3¿ représente Tyr, Asn, Trp ou Phe, A?4¿ représente Lys, Arg ou His, A?5¿ représente Phe, Tyr, Trp, Leu, Val ou Ile et A?6¿ représente Arg, Lys ou His, chaque reste d'acide aminé de cet hexapeptide pouvant être de la forme L ou D, Z représente une chaîne peptidique chargée constituée de restes de 4 à 20 acides aminés de configuration L ou D et Z' représente une chaîne peptidique chargée constituée de restes de 4 à 20 acides aminés de configuration L ou D ou bien est manquant, à condition que Z et Z' ne soient pas tous deux manquants, R¿1? représente H ou un groupe acyle, R¿2? représente NR¿3?R¿4?, R¿3? et R¿4? représentant, de manière indépendante un hydrogène, un alcoxy porteur de 1 à 6 atomes de carbone, un aryloxy ou un alkyle de faible poids moléculaire tel que défini dans le descriptif ou bien R¿2? représente OH. Les conjugats peptidiques correspondant à la formule I peuvent, éventuellement, être, de plus, liés à une fraction de transport. L'invention porte également sur leurs sels, leurs hydrates, leurs solvates et leurs dérivés ayant subi une amidation ou une estérification à la terminaison C avec des acides organiques ou inorganiques appropriés. L'invention concerne, de surcroît, des techniques de production et d'utilisation de ces conjugats. L'invention porte également sur des anticorps se liant de manière spécifique à ces conjugats peptidiques. Le champ d'application de cette invention est très étendu, notamment en matière de traitement d'états pathologiques provoqués par la nociceptine et des peptides d type opioïde connexes.
