An in vitro method is provided herein for detecting the response of intact animal cells to an extra-cellular, exogenous cell activator substance, the activator substance being a substance which when in contact with the intact cells, activates an intracellular enzyme. The method includes contacting the cells in vitro in the presence of a substrate for the activated enzyme with the activator substance immobilised onto a solid support. The consequent activity of the activated enzyme in the presence of the enzyme substrate is then detected. Such contact takes place in one or more wells of a microtiter plate. The well or wells of the microtiter plate, in which the contact takes place between the intact cells, the immobilised activator and the enzyme substrate has or have (each of them) a vertical side wall height of 1 to 2 mm. The method is carried out such that the reaction of the internal diameter of the well (in mm.) to the volume of the sample (in µ1.) is at least 0.1:1, and the ratio of the vertical height of the well to the vertical height (depth) of the sample in the well is in the range 1.1: 1 to 5:1.
