Combination therapies of a cannabinoid receptor agonist and a cannabinoid antagonist at a concentration effective to potentiate, but not antagonize, the therapeutic effect of the cannabinoid receptor agonist are provided. Also provided are methods for use of these combination therapies in potentiating a therapeutic effect of cannabinoid receptor agonists, inhibiting development of acute and chronic tolerance to cannabinoid receptor agonists and treating conditions treatable with cannabinoid receptor agonists in a subject. In addition, a method for reversing cannabinoid receptor agonist tolerance and/or restoring therapeutic action of a cannabinoid receptor agonist in a subject via administration of a cannabinoid receptor antagonist at a concentration effective to potentiate, but not antagonize the therapeutic effect of the cannabinoid receptor agonist is provided.
