Coated microparticles for sustained-release of an active pharmaceutical ingredient after parenteral administration, and methods of producing such coated microparticles, are provided. The coated microparticles comprise core particles of an active pharmaceutical ingredient and a first polymeric coating on the core particles. The first polymeric coating is permeable to the active pharmaceutical ingredient forms a saturated solution within the coated microparticle, resulting in pseudo-zero-sustained release period. A second polymeric coating can be employed to add mechanical support or as a rate- controlling element.
L'invention concerne des microparticules enrobées destinées à la libération prolongée d'un ingrédient pharmaceutique actif après son administration parentérale et des procédés de fabrication de ces microparticules enrobées. Les microparticules enrobées comprennent des particules de noyau d'un ingrédient pharmaceutique actif et un premier revêtement polymère sur les particules de noyau. Le premier enrobage polymère est perméable à l'ingrédient pharmaceutique actif et forme une solution saturée à l'intérieur de la microparticule enrobée, ce qui assure une période de libération pseudo-nulle. Un deuxième enrobage polymère peut s'utiliser pour ajouter un support mécanique ou un élément de régulation de débit.
