A class of 3-phenylimidazo(1,2-a) pyrimidine derivatives (of Formula I, or salt or prodrug thereof : I) wherein Y represents a chemical bond, an oxygen atom, or a -NH- linkage; Z represents an optionally substituted aryl or heteroaryl group; R1 represents hydrogen, hydrocarbon, a heterocyclic group, halogen, cyano trifluoromethyl, nitro, -ORa, -SRa, -SORa, -SO2Ra, -SO2NRaRb,- NRaRb, -NRaCORb, -NRaCO2Rb, -CORa, CO2Ra, -CONRaRb or -CRa=NORb; andRa and Rb independently represent hydrogen, hydrocarbon or a heterocyclic group.), substituted at the meta position of the phenyl ring by an optionally substituted aryl or heteroaryl group which is directly attached or bridged by an oxygen atom or a -NH- linkage, are selective ligands for GABAA receptors, in particular having good affinity for the a2 and/or a3 and/or a5 subunit thereof, and are accordingly of benefit in the treatment and/or prevention of adverse conditions of the central nervous system, including anxiety, convulsions and cognitive disorders.
La présente invention concerne une classe de dérivés de 3-phénylimidazo(1,2-a) pyrimidine (ou un sel ou un promédicament de ceux-ci) (représentés par la formule I dans laquelle: (I)Y représente une liaison chimique, un atome d'oxygène, ou une liaison -NH-; Z représente un groupe aryle ou hétéroaryle éventuellement substitué; R?1¿ représente hydrogène, hydrocarbure, un groupe hétérocyclique, halogène, cyano trifluorométhyle, nitro, -OR?a¿, -SR?a¿, -SOR?a¿, -SO¿2?R?a¿, -SO¿2?NR?a¿R?b¿,-NR?a¿R?b¿, -NR?a¿COR?b¿, -NR?a¿CO¿2?R?b¿, -COR?a¿, CO¿2?R?a¿, -CONR?a¿R?b¿ ou -CR?a¿=NOR?b¿; et R?a¿ et R?b¿ représentent individuellement hydrogène, hydrocarbure ou un groupe hétérocyclique), substitués dans la position méta du cycle phénylique par un groupe aryle ou hétéroaryle éventuellement substitué, directement lié ou ponté par un atome d'oxygène ou une liaison -NH-. Par ailleurs, ces dérivés qui constituent des ligands sélectifs pour les récepteurs GABA¿A?, sont particulièrement caractérisés par une bonne affinité pour la sous-unité a2 et/ou a3 et/ou a5, et par conséquent, sont utiles dans le traitement et/ou la prévention d'états négatifs du système nerveux central, notamment l'anxiété, les convulsions et les troubles cognitifs.
