A process for preparing optically active 2-[6-(hydroxy-methyl)-1,3-dioxan-4- yl]acetic acid derivatives which comprises reacting an enolate prepared by reacting an acetic acid ester derivative with a base or a zero-valent metal with (S)-.beta.-hydroxy-.gamma.-butyrolactone at a temperature of -30~C or above to thereby obtain a dihydroxyoxohexanoic acid derivative, treating this derivative with an acylating agent in the presence of a base to thereby obtain a monoacylated derivative of dihydroxyoxohexanoic acid, reducing the monoacylated derivative with a microorganism into a monoacylated derivative of trihydroxyhexanoic acid, treating the resulting derivative with an acetal- forming reactant in the presence of an acid catalyst to thereby obtain an acyloxymethyldioxanylacetic acid derivative, and subjecting this derivative to solvolysis in the presence of a base.
La présente invention concerne un procédé de préparation de dérivés optiquement actifs d'acide 2-[6-(hydroxy-méthyl)-1,3-dioxan-4-yl] acétique, consistant à soumettre un énolate obtenu par réaction d'un dérivé d'acétate ester avec soit une base, soit un métal à valence nulle, à une réaction avec (S)-.beta.-hydroxy-.gamma.-butyrolactone à -30 ·C ou plus pour un obtenir un dérivé d'acide dihydroxyoxohéxanoïque, puis à traiter ce dérivé avec un agent d'acylation en présence d'une base pour obtenir un dérivé monoacylé d'acide dihydroxyoxohéxanoïque, à réduire ce dérivé monoacylé à l'aide de micro-organismes pour obtenir un dérivé monoacylé d'acide trihydroxyhéxanoïque lequel est traité par un réactif acétalogène en présence d'un acide pour donner un dérivé d'acide acyloxyméthyldioxanylacétique, qu'on soumet enfin à une solvolyse en présence d'une base.
