The invention provides a method for treating cancer in a human comprising (a) administering to the human a dose of a pharmaceutical composition comprising (i) a pharmaceutically acceptable carrier and (ii) an adenoviral vector comprising a nucleic acid sequence encoding a human TNF-a and operably linked to a promoter, wherein the dose comprises about 4 x 10~7 to about 4 x 10~12 particle units (pu) of adenoviral vector, at least once in a therapeutic period comprising up to about 10 weeks, (b) administering a dose of ionizing radiation to the human over the duration of the therapeutic period, and (c) administering a dose of one or more chemotherapeutics to the human over the duration of the therapeutic period, whereby the cancer in the human is treated.
L'invention a pour objet une méthode de traitement du cancer chez un humain consistant à (a) administrer à l'humain d'une dose d'une composition pharmaceutique contenant (i) un excipient pharmaceutiquement acceptable et (ii) un vecteur adénoviral renfermant une séquence d'acides nucléiques codant le facteur onconécrosant-.alpha. humain et liée de manière opérationnelle à un promoteur. La dose renferme entre environ 4 x 10?7¿ et environ 4 x 10?12¿ unités particulaires (up) d'un vecteur adénoviral, au moins une fois dans une période thérapeutique sur au moins environ 10 semaines. La méthode de traitement du cancer consiste également à (b) administrer une dose de rayonnement ionisant sur l'humain pendant la durée du traitement et (c) administrer une dose d'une ou plusieurs chimiothérapies à l'humain pendant la durée du traitement afin de traiter le cancer.
