Provided are modified cellulase enzymes exhibiting increase cellulose-hydrolyzing activity in the presence of lignin and/or reduced binding to lignin comprising modified linker peptides comprising one or more amino acid substitutions, insertions, or deletions that result in (a) a decrease in the calculated isoelectric point of the linker peptide and/or (b) an increase in the ratio of threonine: serine in the linker peptide relative to a parental linker peptide from which said modified linker peptide is derived. Also provided are genetic constructs comprising nucleic acidsequences encoding for modified cellulase enzymes, methods for the production of the modified cellulase enzymes from host strains and a process for hydrolysing cellulose with the modified cellulases in the presence of lignin.
L'invention porte sur des enzymes cellulases modifiées présentant une activité accrue d'hydrolyse de la cellulose en présence de lignine et/ou une fixation réduite à la lignine, lesdites enzymes comprenant des peptides lieurs modifiés comprenant une ou plusieurs substitutions, insertions ou délétions d'acides aminés qui conduisent à (a) une diminution du point isoélectrique calculé du peptide lieur et/ou (b) une augmentation du rapport thréonine/sérine dans le peptide lieur par rapport à un peptide lieur parent à partir duquel ledit peptide lieur modifié est dérivé. L'invention porte également sur des constructions géniques comprenant des séquences d'acides nucléiques codant pour les enzymes cellulases modifiées, sur des procédés de production des enzymes cellulases modifiées à partir de souches hôtes, et sur un procédé d'hydrolyse de cellulose avec les cellulases modifiées en présence de lignine.
