An IgG antibody is provided having a binding affinity for the CD3 antigen complex in which in the heavy chain has a variable region framework together with at least one CDR selected from the amino acid sequences of SEQ ID No 2, 4 and 6 and respective conservatively modified variants thereof and the light chain has a variable region framework together with at least one CDR selected from the amino acid sequences of SEQ ID No 8, 10 and 12 and respective conservatively modified variants thereof characterised in that the heavy chain variable region framework corresponds in sequence to the human type sequence and the light chain variable region framework includes one or more of the specific amino acids characteristic of the rodent type sequence. The novel antibody is capable of being expressed by mammalian cell expression systems at enhanced yields.
Cette invention a trait à un anticorps IgG ayant une affinité de liaison pour le complexe antigénique CD3 dans lequel la chaîne lourde possède une infrastructure de région variable avec au moins une CDR choisie dans le groupe constitué par les séquences aminoacides SEQ ID N· 2, 4 et 6 et leurs variants respectifs raisonnablement modifiés et dans lequel la chaîne légère possède une infrastructure de région variable avec au moins une CDR choisie dans le groupe constitué par les séquences aminoacides SEQ ID N· 8, 10 et 12 et leurs variants respectifs raisonnablement modifiés, se caractérisant par le fait que l'infrastructure de région variable de chaîne lourde correspond, s'agissant de séquence, à une séquence de type humain et que l'infrastructure de région variable de chaîne légère comporte une caractéristique aminoacide spécifique, sinon plusieurs, de la séquence du type rongeur. Ce nouvel anticorps est à même d'être exprimé par des systèmes d'expression de cellule mammalienne et ce, avec un rendement accru.
