Hypermethylation of the SPARC promoter is identified as a mechanism for repressing SPARC gene in cancer resulting in resistance to therapy. The restoration of SPARC expression with demethylating agents, such as 5-Aza-2'deoxycytidine, is shown to enhance chemosensitivity. The invention provides a mechanism of limiting the number of patients exposed to toxicity of demethylating agents by targeting its administration to the subset of patients with hypermethylation of the SPARC promoter.

L'hyperméthylation du promoteur de SPARC est identifiée comme mécanisme pour provoquer la répression du gène SPARC dans un cancer conduisant à une résistance à la thérapie. Il a été démontré que la restauration de l'expression de SPARC par des agents déméthylants, tels que la 5-aza-2'-désoxycytidine, améliore la chimiosensibilité. L'invention propose un mécanisme de limitation du nombre de patients exposés à la toxicité d'agents de déméthylation par ciblage de son administration au sous-ensemble de patients présentant une hyperméthylation du promoteur deSPARC.
