The present invention relates to a glycosylated immunoglobulin or a fragment thereof, in which an immunoglobulin variant, comprising one or more amino acid modifications selected from the group consisting of M160N, A195N, T243N, E265N, Y299T, F331T and Q346N, is additionally glycosylated, and a gene encoding the same. Also, the present invention relates to a glycosylated fusion protein formed as a result of linkage of (a) a glycosylated immunoglobulin or a fragment thereof, in which an immunoglobulin variant having a modified amino acid sequence forming one or more Asn- X-Ser/Thr sequences is additionally glycosylated, with (b) at least one biologically active protein or a portion thereof, a gene encoding the same, a recombination expression vector comprising the gene, a host cell transformed or transfected with the recombinant expression vector, and a method of preparing a glycosylated fusion protein comprising culturing the transformant or transfectant and isolating the glycosylated fusion protein from the culture, and a pharmaceutical composition comprising the glycosylated fusion protein thus prepared.
L'invention concerne une immunoglobuline glycosylée ou un fragment de celle-ci, dans laquelle un variant d'immunoglobuline, comprenant une ou plusieurs modifications d'acide aminé choisies dans le groupe comprenant M160N, A195N, T243N, E265N, Y299T, F331T et Q346N, est glycosylé. L'invention porte également sur : un gène codant pour ces derniers ; et sur une protéine de fusion glycosylée formée par la liaison de (a) une immunoglobuline glycosylée ou un fragment de celle-ci, dans laquelle un variant d'immunoglobuline possédant une séquence aminoacide modifiée formant une ou plusieurs séquences Asn-X-Ser/Thr est en outre glycosylé, avec (b) au moins une protéine active au plan biologique ou une partie de celle-ci ; sur un gène codant cette dernière ; sur un vecteur d'expression recombinant comprenant ledit gène ; sur une cellule hôte transformée ou transfectée avec le vecteur d'expression recombinant. L'invention se rapporte encore à un procédé de préparation d'une protéine de fusion glycosylée, qui consiste à mettre le transformant ou le transfectant en culture, à isoler la protéine de fusion glycosylée du milieu de culture, et à une composition pharmaceutique comprenant la protéine de fusion glycosylée ainsi préparée.
