A DNA isolate is disclosed having a DNA sequence or comprising a fragment of the sequence encoding the protein phosphatase type 1 (PP1) G-subunit which sequence contains a mutation of G to T in the first position of codon 905 such that aspartic acid905 in the expressed PP1 G-subunit is substituted by tyrosine or which fragment spans the mutation. The isolate is useful in the diagnosis of predisposition to insulin resistance or reduced insulin-stimulated non-oxidative glucose metabolism in a subject.
