Methods are provided for substantially reducing background signals encountered in nucleic acid hybridization assays. The method is premised on the elimination or significant reduction of the phenomenon of nonspecific hybridization, so as to provide a detectable signal which is produced only in the presence of the target polynucleotide of interest. In addition, a novel method for the chemical synthesis of isoguanosine or 2'-deoxy-isoguanosine is provided. The invention also has applications in antisense and aptamer therapeutics and drug discovery.

L'invention concerne des procédés de réduction sensible de signaux d'arrière-plan qu'on trouve dans des techniques d'hybridation d'acides nucléiques. Ces procédés se basent sur l'élimination ou la réduction importante du phénomène d'hybridation non spécifique, de manière à produire un signal détectable généré uniquement en présence du polynucléotide cible concerné. L'invention concerne, de plus, un nouveau procédé de synthèse chimique d'isoguanosine ou de 2'-deoxy-isoguanosine. L'invention peut également être mise en application dans des thérapies à base de molécules antisens et d'aptamères, ainsi que dans la recherche de médicaments.
