A method for determining the sequence of a polynucleotide comprising the steps of (i) contacting a polynucleotide processive enzyme immobilised in a fixed position, with a target polynucleotide under conditions sufficient to induce enzyme activity; (ii) detecting an effect consequent on the interaction of the enzyme and the polynucleotide, wherein the effect is detected by measurement of a non-linear optical signal or a linear signal coupled to a non-linear signal.

L'invention concerne un procédé destiné à déterminer la séquence d'un polynucléotide qui consiste: (i) à mettre en contact une enzyme de maturation polynucléotidique immobilisée dans un emplacement fixe, avec un polynucléotide cible dans des conditions suffisantes à induire l'activité enzymatique; (ii) à détecter un effet résultant de l'interaction de l'enzyme et du polynucléotide, cet effet étant détecté par mesure d'un signal optique non linéaire ou d'un signal linéaire couplé à un signal non linéaire.
