Novel mammalian phospholipase (PLA2) nucleotide sequences and low molecular weight (about 14 KD) amino acid sequences encoded thereby are disclosed. More particularly, a cloned human HPLA2 cDNA expressing HPLA2 -10 and its cloned rat RPLA2 cDNA counterpart, expressing RPLA2 -10, which are characterized as PLA2 Type IV, are disclosed. A second type of PLA2 cDNA, characterized as PLA2 Type III and exemplified by a rat PLA2 cDNA encoding RPLA2 -8 and a partial human PLA2 nucleotide sequence encoding HPLA2 -8, is disclosed. Expression of the cDNAs encode the two new types of PLA2 enzymes which have phospholipase activity. The novel PLA2s do not include disulfide bridges between cysteine amino acids 11 and 77 or elapid loops. However, the novel PLA2s may include amino acid COOH-terminal extensions which can vary in length. Seventeen of the eighteen absolutely conserved amino acids in all active 14 KD PLA2s are believed to be conserved in RPLA2 -8 and HPLA2 -8, whereas all eighteen are believed to be conserved in RPLA2 -10 and HPLA2 -10. Because me encoded sequences of RPLA2 -8 and HPLA2 -8 include only 16 cysteine amino acids, they have been designated as Type III. RPLA2 -10 and HPLA2 -10 are designated as Type IV since their encoded sequences include only 12 cysteine amino acids.

L'invention concerne de nouvelles séquences nucléotidiques de phospholipases A2 (PLA2) mammifères et des séquences d'acides aminés de faible masse moléculaire (environ 14 KD) codées par ces séquences. Elle porte en particulier sur l'ADNc d'une phospholipase humaine (HPLA2) clonée exprimant HPLA2-10 et sur l'ADNc correspondant d'une phospholipase de rat (RPLA2) clonée, exprimant RPLA2-10 et caractérisés en tant que PLA2 de Type IV. Elle concerne aussi un second type d'ADNc de PLA2, caractérisé en tant que PLA2 de Type III, et dont l'ADNc de PLA2 de rat codant RPLA2-8 est un exemple, ainsi qu'une séquence nucléotidique partielle de PLA2 humaine codant HPLA2-8. Les expressions des ADNc codent les deux nouveaux types d'enzymes de PLA2 ayant une activité de phospholipase. Les nouvelles PLA2 ne comportent pas de ponts disulfure entre les acides aminés de cystéine 11 et 77 ou les boucles d'élapidé. Cependant, les nouvelles PLA2 peuvent contenir des extensions aminoacides COO-H terminal qui peuvent varier en longueur. On pense que seize des dix-huit acides aminés parfaitement conservés dans toutes les PLA2 actives de 14 KD sont conservés dans RPLA2-8 et HPLA2-8, tandis que tous les dix-huit seraient conservés dans RPLA2-10 et HPLA2-10. Du fait que les séquences codées de RPLA2-8 et HPLA2-8 ne contiennent que 16 acides aminés de cystéine, elles sont dites de Type III. RPLA2-10 et HPLA2-10 sont considérées comme étant de Type IV, leurs séquences codées ne contenant que 12 acides aminés de cystéine.
