The present invention relates to chimeric molecules comprising an oligonucleotide complementary to a region of the ribonucleotide component of telomerase attached to an activator of RNase L ("activator-antisense complex") which specifically cleaves the ribonucleotide portion of a telomerase enzyme. The present invention relates to methods of inhibiting telomerase enzymatic activity with activator-antisense complexes targeted to the RNA component of telomerase. The present invention further relates to methods of treating malignant neoplastic disease, wherein the malignant cells contain a telomerase activity that is necessary for the growth of the malignant cells.
L'invention concerne des molécules chimères qui comprennent un oligonucléotide complétant une région de l'élément ribonucléotidique de la télomérase lié à un activateur de la RNase L (complexe antisens activateur) qui clive spécifiquement la partie ribonucléotidique d'une enzyme télomérase. L'invention concerne en outre des procédés d'inhibition de l'activité enzymatique de la télomérase au moyen de complexes antisens activateurs ciblant l'élément ARN de la télomérase. L'invention concerne enfin des méthodes de traitement d'affections néoplastiques malignes dans lesquelles les cellules malignes présentent une activité de télomérase nécessaire pour la croissance desdites cellules malignes.
