The present invention discloses methods for the detection and identification of Kaposi's sarcoma-associated herpesvirus-like Virus (KSHV)-specific nucleic acid sequences or proteins in biological samples derived from patients diagnosed with multiple myeloma (MM) or monoclonal gammopathy of undetermined significance (MGUS). KSHV-specific nucleic acid sequences are detected in these patients by the utilization of polymerase chain reaction (PCR) or reverse transcription polymerase chain reaction (RT-PCR) to amplify viral- specific sequences indigenous to the tissue/cells of theses aforementioned patients. The present invention also discloses several methods for the prophylactic and/or therapeutic treatment of patients with MM or MGUS. Potential treatment modalities disclosed in the present invention include, but are not limited to: (1) the use of inhibitory nucleic acids (INAs), such as "anti-sense" oligonucleotides, to inhibit various viral processes (e.g., DNA replication, mRNA transcription, and/or protein translation); (2) the use of viral-specific polyclonal or monoclonal antibodies, or biologically-active fragments or derivatives thereof, the binding of which may be used to inhibit initial viral infection, replicative/protein synthetic processes, or for the "delivery" of anti-viral agents conjugated to a portion of the immunoglobulin; (3) the utilization of systemic anti-viral chemotherapeutic agents which inhibit viral infection or replicative/protein synthetic processes; and/or (4) the use of a KSHV-specific vaccine to inoculate individuals and elicit a prophylactic or therapeutic immune responses through the utilization of KSHV- specific nucleic acid sequences or recombinant proteins as inoculates to elicit such immune response(s).
La présente invention concerne des méthodes de détection et d'identification de séquences d'acides nucléiques ou de protéines spécifiques à un virus semblable à l'herpèsvirus associé au sarcome de Kaposi (KSHV) dans des échantillons biologiques prélevés sur des patients chez qui on a diagnostiqué un myélome multiple (MM) ou une gammapathie monoclonale de signification non déterminée (MGUS). On détecte les séquences d'acide nucléique spécifiques de KHSV chez ces patients par amplification en chaîne par polymérase (PCR) ou amplification en chaîne par polymérase à transcription inverse (RT-PCR) pour amplifier des séquences spécifiquement virales endogènes des tissus/cellules de ces patients. La présente invention concerne également plusieurs méthodes de traitement prophylactique et/ou thérapeutique de ces patients atteints de MM ou MGUS. Un traitement potentiel selon la présente invention comprend, de manière non exhaustive: (1) l'utilisation d'acides nucléiques inhibiteurs (INAs), tels que des oligonucléotides "antisens" pour inhiber plusieurs processus viraux (comme par exemple une réplication de l'ADN, une transcription d'ARN messager et/ou une traduction de protéine); (2) l'utilisation d'anticorps polyclonaux ou monoclonaux spécifiquement viraux, ou de leurs fragments ou dérivés biologiquement actifs, dont la liaison peut servir à inhiber une infection virale initiale, des processus réplicatifs/de synthèse protéique, ou à "fournir" des agents antiviraux conjugués à une partie de l'immunoglobuline; (3) l'utilisation d'agents chimiothérapeutiques antiviraux systémiques inhibant une infection virale ou des processus réplicatifs/de synthèse protéique; et/ou (4) l'utilisation d'un vaccin spécifique de KHSV inoculé à des individus pour éliciter une réponse immunitaire prophylactique ou thérapeutique par utilisation de séquences d'acides nucléiques ou de protéines recombinées spécifiques de KHSV pour éliciter une (des) réponse(s) immunitaire(s).
