A process for producing motor neurons and GABAergic neurons characterized by comprising suspension-culturing embryonic stem cells in the presence or absence of a protein noggin to form embryoids, deriving the embryoids into nerve stem cells by suspension culture in the presence of a fibroblast growth factor and a sonic hedgehog protein, and then differentiating the same. According to this method, at least motor neurons and GABAergic neurons can be systematically and efficiently produced from ES cells. Selective acquisition of neurons would be applicable to the transplant therapy for amyotrophic lateral sclerosis, Huntington's chorea, Alzheimer's disease, etc.
L'invention porte sur un procédé de production de neurones moteurs et de neurones GABAergiques consistant à produire une culture d'une suspension de cellules souches d'embryons en présence ou non d'une petite quantité de protéines pour former des embryoïdes, puis à transformer les embryoïdes en cellules souches de nerfs par culture d'une suspension en présence du facteur de croissance du fibroblaste et de protéines shh, puis à y effectuer une différenciation. Ledit procédé permet de produire systématiquement et efficacement au moins des neurones moteurs et des neurones GABAergiques à partir de cellules souches d'embryons. Cette acquisition sélective de neurones s'applique au traitement par transplantation, de la sclérose amyotrophique latérale, de la chorée de Huntington, de la maladie d'Alzheimer, etc.
