The present invention relates to a variant of Lkn-1(shLkn-1) with enhanced biological activity, which is a truncated form of Lkn-1, a process for preparing a recombinant shLkn-1 by employing expression vector therefor and pharmaceutical application of the said protein. shLkn-1 is generated by missing 26 amino acid residues from the amino terminus of Lkn-1 to contain 66 amino acids. Recombinant shLkn-1 inhibits colony formation and cell proliferation in vivo, which suggests that it can be used as a potential drug for the antibody production, the treatment during HIV-1 infection, the protection of bone marrow stem cells during chemotherapy or radiotherapy, and the inhibition of leukemia.
Cette invention se rapporte à un variant de Lkn-1(shLkn-1) avec activité biologique accrue, qui est une forme tronquée de Lnk-1, à un procédé de préparation d'un shLkn-1 de recombinaison par utilisation d'un vecteur d'expression pour cette protéine et à une application pharmaceutique de cette protéine. La protéine shLkn-1 est produite par élimination de 26 restes d'acides aminés de la terminaison amino de Lkn-1 afin que cette protéine contienne 66 acides aminés. La shLkn-1 de recombinaison empêche la formation de colonies et la prolifération cellulaire <i>in vivo</i>, ce qui suggère qu'elle puisse être utilisée comme médicament potentiel en vue de la production d'anticorps, le traitement des infections à VIH-1, la protection des cellules souches de moelle osseuse pendant une chimiothérapie ou une radiothérapie et l'inhibition de la leucémie.
