An apparatus and method for transdermal molecular delivery, comprises a first electrode assembly having an anode and a cathode in closely spaced relation for engaging the stratum corneum through which to apply an electric field, a second electrode assembly spaced from the first electrode assembly through which to apply an electric field and comprising at least one of an anode and a cathode, a first power supply including a first circuit preferably connected to the first electrode assembly for applying a pulsed electric field of sufficient amplitude to induce pores in the stratum corneum, second power supply including a second circuit connected to the first electrode assembly and the second electrode assembly for applying a low voltage continuous electric field of a preselected polarity and sufficient amplitude to induce migration of molecules through pores in the stratum corneum. The iontophoresis current density to the electrode assembly is around 0.5 mA per cm2, and after an electroporation pulsing event, between around 2.9 and 7.0 mA per cm2.
L'invention concerne un appareil et un procédé destinés à l'apport transdermique de médicaments, l'appareil comprenant ce qui suit: un premier ensemble d'électrodes possédant une anode et une cathode faiblement espacées qui servent à entrer en contact avec la couche cornée à travers laquelle un champ électrique est appliqué; un deuxième ensemble d'électrodes espacé du premier, à travers lequel est appliqué un champ électrique, comprenant au moins une anode et une cathode; une première alimentation comprenant un premier circuit connecté, de préférence, au premier ensemble d'électrodes et servant à appliquer un champ électrique pulsé ayant une amplitude suffisante pour créer des pores dans la couche cornée; une deuxième alimentation comprenant un deuxième circuit connecté aux premier et deuxième ensembles d'électrodes pour appliquer un champ électrique permanent à faible tension qui possède une polarité présélectionnée et une amplitude suffisante pour provoquer la migration des molécules à travers les pores dans la couche cornée. La densité du courant de l'ionophorèse vers l'ensemble d'électrodes est égale à environ 0,5 mA par cm?2¿ et, après l'événement d'électroporation pulse, environ entre 2,9 et 7,0 mA par cm?2¿.
