A system for rapidly determining the presence or quantity of coliform bacteria in a water sample using the enzymes .beta.-D-galactosidase and .beta.-D- glucuronidase. The system includes a first filter means for separating bacteria from the sample and a broth for culturing the bacteria including an inducing agent for inducing enzyme production. A second filter means is used to separate the cultured bacteria from the broth. A lysing agent is exposed to the bacteria on the second filter and incubated with a chemiluminogenic substrate of the enzyme to produce a chemiluminescent product. Light emission is initiated from the second filter means and the emitted light is detected or measured directly from the second filter means using a luminometer adapted to receive the second filter means. The system is especially effective at improving the sensitivity and specificity of the assay by increasing the signal received from encapsulated target organisms and reducing the interference from non-target organisms that may be present in the sample.

L'invention concerne un système permettant de déterminer rapidement la présence ou la quantité de bactéries coliformes dans un échantillon d'eau au moyen des enzymes .beta.-D-galactosidase et .beta.-D-glucuronidase. Ce système comprend un premier moyen filtrant destiné à séparer les bactéries contenues dans ledit échantillon et un milieu liquide pour la culture des bactéries, contenant un agent inducteur destiné à induire la production d'enzymes. Un second moyen filtrant est utilisé pour séparer les bactéries cultivées contenues dans ledit milieu liquide. Un agent de lyse est exposé aux bactéries sur le second filtre et incubé avec un substrat chimiluminogène de l'enzyme pour former un produit chimiluminescent. L'émission de lumière est initiée à partir du second moyen filtrant et la lumière émise est détectée ou mesurée directement à partir du second moyen filtrant à l'aide d'un luminomètre conçu pour recevoir ce second moyen filtrant. Le présent système améliore de façon particulièrement efficace la sensibilité et la spécificité de l'analyse, en augmentant le signal émis par des organismes cibles encapsulés et en réduisant l'interférence causée par des organismes non cibles éventuellement présents dans l'échantillon.
