Apparatus and methods are disclosed for maturing an elongate replacement tissue construct in vitro prior to use of the replacement construct in vivo as, for example, a ligament. The tissue is seeded with specific cells, exposed to a maturation fluid, and subjected to selected forces, which can include longitudinal stress (i.e. stressing the tissue along its elongate axis). The tissue is disposed in a maturation chamber that confines maturation fluid for introduction to the tissue. A first mounting element couples to a first end of the elongate biopolymer tissue and a second mounting element couples to a second end of the tissue such that the tissue extends along a longitudinal axis, and a force is applied to at least one of the mounting elements for longitudinally stressing the tissue. The foregoing apparatus and methods are intended to provide a replacement tissue that is more readily integrable in vivo, i.e., a tissue that more readily degrades, regenerates and remodels in vivo to produce a more durable and functional replacement tissue.

L'invention concerne un appareil et des méthodes de maturation d'une construction de tissus de remplacement allongés in vitro avant l'utilisation de la construction de remplacement in vivo par exemple, comme ligament. Le tissu est ensemencé avec des cellules spécifiques, il est exposé à un fluide de maturation et soumis à des forces choisies, lesquelles peuvent comprendre un effort longitudinal (c'est-à-dire que l'on soumet le tissu à un effort le long de son axe longitudinal). Le tissu est disposé dans une chambre de maturation contenant un fluide de maturation destiné à être introduit dans le tissu. Un premier élément de support se couple à une première extrémité du tissu allongé en biopolymère et un second élément de support se couple à une seconde extrémité du tissu de manière que le tissu s'étende le long d'un axe longitudinal, et une force est appliquée à au moins des éléments de support pour soumettre le tissu à un effort longitudinal. L'appareil et les méthodes de l'invention sont destinés à produire un tissu de remplacement plus facilement intégrable in vivo, c'est-à-dire un tissu se dégradant, se régénérant et se remodelant plus facilement in vivo afin de produire un tissu de remplacement plus durable et plus fonctionnel.
